One woman has died and two more have been taken to hospital after a serious car accident south of Adelaide.

Police and emergency crews were called to the intersection of McLaren Flat and Baker Gully roads, near Kangarilla, at 9am this morning after a four-wheel-drive towing a caravan and a Honda sedan collided.

A 72-year-old man and a 68-year-old woman from Aldinga Beach who were traveling in the four-wheel-drive were airlifted to the Flinders Medical Centre with serious injuries.

The driver of the sedan was killed. She was the vehicle's only occupant.

Senior sergeant Brenden Kenny from South Coast Police said the tragedy should serve as a warning to other drivers as a long weekend approaches.

"This is a scene that's been replicated too many times, particularly in this last week," he said.

"What we would ask of our motorists, particularly as we go towards the long weekend next weekend, is that they exercise a great amount of care; driving is one of the most dangerous things that people can do."

Road closures are in place and motorists have been asked to avoid the area.
