Police say the man allegedly threatened staff at the Grand Junction Rd chemist at about 9.20am and stole the till.
Staff followed the man on foot for a short time after he left the premises.
No-one was injured and while the stolen till has been recovered, police are still looking for the alleged offender.
He is described as Caucasian, about 30-years-old, with a bald or shaved head, wearing a black leather jacket, khaki trousers and black fingernails.RT @7Sport_au: Essendon's Lovett-Murray stabbed | http://t.co/lIbeOPeOSU #AFL #essendon
